Extended field-of-view light-sheet microscopy

چکیده
Light-sheet fluorescence microscopy enables rapid 3D imaging of biological samples. Unlike confocal and two-photon microscopes, a light-sheet microscope illuminates the focal plane with an objective orthogonal to detection axis images it in single snapshot. Its combination high contrast minimal sample exposure make ideal image thick samples sub-cellular resolution. To uniformly illuminate wide field-of-view without compromising axial resolution, propagation-invariant light-fields such as Bessel Airy beams have been put forward. These do however irradiate relatively broad transversal structure. The excited by side lobes can be blocked physically during recording; though at cost increased exposure. In contrast, beam has fine structure that is both curved asymmetric. captures all high-frequency components enable resolution need discard useful fluorescence. This advantage does not carry over naturally excitation where suppressed. We demonstrate symmetric planar used rely on deconvolution.

Pupil filters for extending the field-of-view in light-sheet microscopy.
Pupil filters, represented by binary phase modulation, have been applied to extend the field of view of a light-sheet fluorescence microscope. Optimization has been used, first numerically to calculate the optimum filter structure and then experimentally, to scale and align the numerically synthesized filter in the microscope. A significant practical extension of the field of view has been obse...

Increasing the volumetric imaging speed of light-sheet microscopy will improve its ability to detect fast changes in neural activity. Here, a system is introduced for brain-wide imaging of neural activity in the larval zebrafish by coupling structured illumination with cubic phase extended depth-of-field (EDoF) pupil encoding. This microscope enables faster light-sheet imaging and facilitates a...
متن کاملLight-sheet microscopy with digital Fourier analysis measures transport properties over large field-of-view.
Using light-sheet microscopy combined with digital Fourier methods we probe the dynamics of colloidal samples and DNA molecules. This combination, referred to as selective-plane illumination differential dynamic microscopy (SPIDDM), has the benefit of optical sectioning to study, with minimal photobleaching, thick samples allowing us to measure the diffusivity of colloidal particles at high vol...
متن کاملHyperspectral light sheet microscopy
To study the development and interactions of cells and tissues, multiple fluorescent markers need to be imaged efficiently in a single living organism. Instead of acquiring individual colours sequentially with filters, we created a platform based on line-scanning light sheet microscopy to record the entire spectrum for each pixel in a three-dimensional volume. We evaluated data sets with varyin...
متن کاملConfocal multiview light-sheet microscopy.
Selective-plane illumination microscopy has proven to be a powerful imaging technique due to its unsurpassed acquisition speed and gentle optical sectioning. However, even in the case of multiview imaging techniques that illuminate and image the sample from multiple directions, light scattering inside tissues often severely impairs image contrast. Here we combine multiview light-sheet imaging w...
